SAMPLES WITH 1, 2, AND 5 G/L TX-100 GAVE GOOD SPECTRA, AS JUDGED BY FITTING WITH LAST.
Actually since vol was only .6 ml, these were 1.67, 3.3 and 8.3 g/L TX-100
good heme spectra in 25, 30, 41
subtract 44 for baseline
Imidazole buffer w no protein has strong absorbance but at shorter wavelength.
Use ave(40,41,42) vs 43,44 for spectrum of hemochrome;
43,44 vs 45,46 for spectrum of color in imidazole.
Fit all the spectra with this, the three highest TX-100 concentrations fit excellently
lower concentrations do not.
Lets say use 5 g/l TX-100 in assay mix. For huge amount of protein, may need to go higher.
